Evaluación del potencial antiinflamatorio de fotoprotectores mediante acoplamiento molecular sobre COX-2 y 5-LOX
Anti-inflammatory potential of photoprotective compounds: a molecular docking analysis against COX-2 and 5-LOX

La radiación ultravioleta (UV) puede desencadenar procesos inflamatorios cutáneos asociados a la producción de mediadores derivados del ácido araquidónico por medio de las enzimas ciclooxigenasa-2 (COX-2) y 5-lipoxigenasa (5-LOX). En este estudio preliminar, se evaluó mediante acoplamiento molecular el potencial antiinflamatorio de 11 compuestos con actividad fotoprotectora conocida y 4 polifenoles frente a ambas dianas enzimáticas. A partir de las puntuaciones de fitness, se priorizaron los ligandos con mejor desempeño relativo dentro de la serie. Los polifenoles no exhibieron características farmacofóricas compatibles. Frente a COX-2, solo el iscotrizinol (87,86) y el DHHB (71,64) mostraron puntuaciones superiores a la del sustrato natural, el ácido araquidónico (AA), con un score de 70,53. En comparación con 5-LOX, se identificó un perfil similar al observado para COX-2 con el iscotrizinol (70,59), así como un comportamiento distinto caracterizado por la tendencia a mayores puntuaciones de fitness en compuestos voluminosos y lipofílicos, como bemotrizinol (72,07) y bisoctrizole (70,01), que alcanzaron puntuaciones próximas o superiores a la del AA (70,95). Este estudio puso de relieve que valores elevados de fitness score no siempre corresponden a modos de unión estructuralmente favorables, lo que permitió identificar falsos positivos asociados al efecto del tamaño molecular o a una acomodación inadecuada, como se observó para iscotrizinol, bisoctrizole, bemotrizinol y DHHB. En contraste, el octocrileno-R presentó interacciones consistentes frente a COX-2, mientras que el salicilato de octilo fue el único ligando que mostró un perfil de interacción estructuralmente coherente frente a ambas enzimas. En conjunto, los datos sugieren que algunos compuestos con actividad fotoprotectora pueden interactuar con dianas enzimáticas implicadas en la inflamación; sin embargo, este potencial debe ser confirmado mediante estudios experimentales y simulaciones adicionales.
