Potencial anti-inflamatório de fotoprotetores: uma análise de docking molecular em COX-2 e 5-LOX
Anti-inflammatory potential of photoprotective compounds: a molecular docking analysis against COX-2 and 5-LOX

A radiação ultravioleta (UV) pode desencadear processos inflamatórios cutâneos associados à produção de mediadores derivados do ácido araquidônico por meio das enzimas ciclooxigenase-2 (COX-2) e 5-lipoxigenase (5-LOX). Neste estudo preliminar, avaliou-se, por docking molecular, o potencial anti-inflamatório de 11 compostos com conhecida atividade fotoprotetora e 4 polifenóis, frente aos dois alvos enzimáticos. A partir das pontuações de fitness foram priorizados ligantes com melhor desempenho relativo, dentro da série. Frente à COX-2, apenas o iscotrizinol (87,86) e o DHHB (71,64) exibiram pontuação superior à do substrato natural, o ácido araquidônico (AA), de score 70,53. Frente à 5-LOX, o iscotrizinol (70,59) apresentou perfil semelhante ao observado para a COX-2. Um comportamento distinto foi revelado pela tendência de maiores pontuações fitness para os compostos volumosos e lipofílicos: bemotrizinol (72,07) e bisoctrizol (70,01) que atingiram pontuações próximas ou superiores à do AA (70,95). Este estudo ressaltou que valores elevados de fitness score nem sempre correspondem ao modo de ligação estruturalmente favorável, o que permitiu a identificação de falsos positivos associados ao efeito de tamanho ou à acomodação inadequada, como observado para iscotrizinol, bisoctrizol, bemotrizinol e DHHB. O (R)-octocrileno, por sua vez, apresentou interações consistentes frente à COX-2, enquanto o salicilato de octila foi o único ligante a exibir perfil de interação estruturalmente coerente frente a ambas as enzimas. Os dados sugerem que alguns compostos com atividade fotoprotetora podem interagir com alvos enzimáticos envolvidos na inflamação, entretanto esse potencial deva ser confirmado por estudos experimentais e simulações adicionais.
